Appendix A. Protocolonderbreking: opslag en het hertesten van monsters
1. Om een warmtebehandeld lysaat te bewaren, sluit u het lysebuisje opnieuw af met een schone dop (zie hoofdstuk
4.5, Lyse).
2. Incubeer een verrijkt monster gedurende minimaal 18 uur voorafgaand aan de opslag.
3. Sla gedurende maximaal 72 uur op bij een temperatuur van 4 tot 8 °C.
4. Bereid een opgeslagen monster voor op de amplificatie door het 2-3 keer om te keren om het te mengen.
5. Verwijder de doppen van de buisjes.
6. Plaats de gemengde lysaatbuisjes op het 3M Moleculaire Detectie - Verwarmingsblokinzetstuk en verwarm
gedurende 5 ± 1 minuten op een temperatuur van 100 ± 1 °C.
7. Haal het rek met buisjes met 3M Lyse-oplossing uit het verwarmingsblok en laat het minimaal 5 minuten en maximaal
10 minuten afkoelen in het 3M Moleculaire Detectie - Koelblokinzetstuk.
8. Ga verder met het protocol zoals hierboven in het hoofdstuk Amplificatie beschreven.
