Annexe A. Interruption du protocole : stockage et nouveau test des lysats soumis à un traitement thermique
1. Pour conserver un lysat soumis à un traitement thermique, refermer le tube de lyse avec un bouchon propre (voir
«Lyse», 4.5)
2. Stocker à une température comprise entre 4 et 8 °C jusqu'à 72 heures.
3. Préparer un échantillon conservé pour amplification en retournant 2 à 3 fois pour mélanger.
4. Ouvrir les tubes.
5. Placer les tubes de lysats mélangés dans le Support de bloc chauffant pour système de détection moléculaire 3M et
chauffer à 100 ± 1 °C pendant 5 ± 1 minute.
6. Retirer le portoir non couvert de tubes de LS du bloc chauffant et le laisser refroidir dans le Support de bloc refroidissant
pour système de détection moléculaire 3M entre 5 et 10 minutes.
7. Poursuivre le protocole à la section « Amplification » détaillée ci-dessus.
