8.Plan de travail pour realiser une
lyophilisation
For this proceed as follows:
/ Close the shutoff valve (32).
After this:
/ Press the DRYING button (51).
O The vacuum pump (16) is switched on after a time de-
lay and the LED lights up.
e) Attaching the bulbs
As soon as a pressure between 5 x 1O
2
to 1 x 10-' mbar has
been reached, it is possible to attach the bulbs filled with
frozen product.
To do this proceed as follows:
/ Open the shutoff valve (32).
/ Attach the first bulb with frozen product to the plastic
cone of one of the manifold plate valves (23).
As soon as the bulb has been mounted correctly on the
cone:
</ Open the valve by turning the swivelling lever 180° so
that the nose is pointing down.
/ Attach the other bulbs at appropriate intervals (15 to 20
minutes) as described above.
Note: .
The individual bulbs should be attached at intervals of
at least 15 minutes so that stable drying conditions can
settle in each of the individual bulbs.
The operating pressure will increase depending on the
number of bulbs that have been attached. To prevent the
product from melting, therefore, the pressure should be
checked before attaching each bulb.
f) Ending the freeze-drying process
During the drying process, moisture from the air con-
denses on the outer surfaces of the bulbs. In general, one
can assume that the product is dry when the following con-
ditions have been met:
O The ice on the outer surfaces has melted.
O The bulbs have reached ambient temperature.
To check whether this is the case:
/ Touch the outer surface of the bulb with the palm of
your hand.
As soon as the drying process has ended, proceed as
follows:
/ Close the valves (23) by turning the swivelling levers
180° so that their noses point up.
/ Vent the bulbs.
To do this proceed as follows:
• Hold the bulb firmly at the bottom and remove it by
pulling down from the corresponding valve of the
manifold plate.
As soon as all the bulbs have been removed:
/ Wail approximately 30 minutes and then press the
DRYING button (51) to switch the unit off.
La vanne d'isolation fermee (position ,,ZU") ensuite:
/ appuyer sur la touche ,,DRYING" (51)
O la pompe a vide (16) fonctionne et la touche s'illumine
e) Disposer les ballons:
Lorsque la pression se situe entre 5 x 10

mbar et 1 x 10-'
mbar on peut disposer les ballons congeles sur la rampe de
sechage (un par un)
/ la vanne d'isolation (32) ouverte et
</ disposer le premier ballon avec produit congele au pre-
mier robinet conique de la rampe de sechage.
Lorsque le ballon est bien introduit dans le cone, imme-
diatement:
/ tourner le bee du robinet de 180° (bee vers le bas) et le
robinet s'ouvre
</ la pression remonte alors et Ton attend que la pression
de travail s'etablisse vers les 5 x 10-' mbar pour mettre
le second ballon et ainsi de suite.
Remarque:
Entre chaque disposition de ballon, il s'ecoule un temps
dc minimum 15 minutes, celui-ci est necessaire pour
rctablir la pression clans l'enceinte et eviter une decon-
gelation du produit contenue dans le ballon.
Independammcnt du nombrc de ballons a disposer, il
est necessaire de bien surveiller la pression de travail et
d'eviter dc disposer de nouveau ballon a pression trop
elevee ce qui aurait pour effet de decongeler le produit
conlenu dans les ballons.
f) Fin de lyophilisation
Le ballon adapte sur le robinet est givre a Pexterieur, et
Ton peut affirmer que le produit est lyophilise lorsque:
O le givre exterieur au ballon a disparu et que le ballon a
atteint la temperature ambiante.
Vous pouvez controler en testant avec la main la t° ex-
terieure du ballon qui devrait etre egale a la t° ambiante.
Lorsque la lyophilisation est terminee:
/ les robinets sont mis en position de remise a Pair par ro-
tation des bees de 180° (bee vers le haut)
Ensuite :
/ enlever les ballons de leur cone d'adaption
Lorsque tous les ballons sont enleves:
/ appuyer sur la touche ,,DRYING" (51) apres 30 minu-
tes et l'appareil s'arrete de fonctionner.
