5.C. Two-stage Purifi cation
1. Confi gure the system as shown in Figure 2.
2. Close all valves (V1 - V8).
3. Position V8 to direct fl ow to the inlet.
4. Open all vent valves (V2, V3, V5 and V6).

IMPORTANT NOTICE: 3M Zeta Plus Capsules: Pre-fl ushing the system with 54 L/m
buff er or water is required. Maximum pre-fl ushing fl ux is 300 Liters/m
Emphaze AEX Hybrid Purifi er Capsules: The capsules must be fl ushed prior to use to remove
the glycerin that is present as a media stabilizer. Flush the capsule with 15.3 mL/bed volume
of buff er or 25-150 mm sodium chloride solution. Do not use water as the fl ush solution.
5. Open V1 slowly to begin fl uid fl ow.
6. When the fl uid exits V3, close V3.
7. When fl uid exits V2, close V2 and simultaneously position V4 to direct the fl uid to V8.
8. When fl uid exits V6, close V6.
9. When fl uid exits V5, close V5 and simultaneously open V7.
10. Close V1 and then close V7 to stop fl ow.
11. Drain fl uid from the system if required by turning V1, V4, V7 and V8 to the drain position. Then
open V2, V3, V5, and V6.
12. Filter process fl uid by repeating steps 2 – 10 with process fl uid.
To reduce the risk of Injury from possible end user exposure associated with
degradation products released from the purifi cation media; and
To reduce the risk of product loss associated with a) chemical release from
purifi cation media and b) nullifi cation of Supplier's USP<88> Class VI compliance of
fi lter capsule components.
• The capsule must be subjected to a preconditioning fl ush prior to use.
IMPORTANT NOTICE: DO NOT blow down 3M Emphaze AEX Hybrid Purifi er capsules after
pre-conditioning fl ush procedure.
IMPORTANT NOTICE: For optimal performance, run fi ltration process at a fl ux of 160-630
LMH (0.75-3.0 Bed Volume per minute).
13. At the end of fi ltration, the fl uid remaining on the upstream side may be fi ltered through the
system:
a. Open V3 and reduce the system pressure to zero (0) psi (0 bar).
b. Apply a maximum of 20 psi (1.4 bar) air pressure at a temperature less than 25°C to valve V3.
c. Monitor the system outlet. When liquid fl ow is reduced to a trickle from the 1st Stage capsule,
reduce the system pressure to zero (0) psi (0 bar) and then close V3 and V8.
d. Open valve V6 and apply a maximum of 20 psi (1.4 bar) air pressure at a temperature less than
25°C to blow down the capsules post use.
e. Monitor the system outlet. When liquid fl ow is reduced to a trickle from the 2nd Stage
capsule, reduce the system pressure to zero (0) psi (0 bar) and then close V6 and V8.
