4.C. Single Stage Purifi cation Instructions
The Production Capsule must be fl ushed prior to use to remove the glycerin that is present as a
media stabilizer. Flush the capsule with 54 L/m
having a conductivity in the
same range as that of 25 to
150 mM NaCl (3 - 16 mS/cm)
at a fl ow rate no higher than
210 L/m
2
/h (LMH). DO NOT
use water as the fl ush solution.
Flush in the forward fl ow
direction only. Intermittently
bleed air off using the Air Vent
on the manifold.
IMPORTANT NOTICE:
Precondition fl ush the
capsule prior to use.
4.C.1 Required
Preconditioning Flush Procedure
1. Connect the inlet and outlet located on the bottom manifold to the upstream and downstream
process equipment.
IMPORTANT NOTICE: Do not rotate the production-scale holder when process equipment
is attached.
2. Connect vent valves to the upstream and downstream vent ports located on the top manifold.
3. Connect the capsule inlet to the reservoir containing the pre-conditioning fl ush solution.
4. Open both vent valves.
5. Close the outlet valve and open the inlet valve slowly to begin the fl ow of
pre-conditioning fl ush solution through the inlet valve.
6. When fl uid emerges from the upstream vent valve, close the upstream vent valve.
7. When fl uid emerges from the downstream vent valve, close the valve while simultaneously
opening the outlet valve slowly.
8. Pump the appropriate amount of fl ush solution per capsule size to complete the required
preconditioning fl ush.
9. Close the inlet valve and stop the fl ow of pre-conditioning fl ush solution.
IMPORTANT NOTICE: Do not blow down 3M
pre-conditioning fl ush procedure.
IMPORTANT NOTICE: For optimal performance, run fi ltration process at a fl ux of 160 – 630
LMH (0.75-3.0 Bed Volume per minute).
4.C.2. Single-stage Purifi cation
1. Connect the inlet and outlet of the capsule to the upstream and downstream process
equipment, respectively.
2. Connect the capsule inlet to the reservoir containing the bioprocess fl uid.
3. Start the fl ow of bioprocess fl uid and collect purifi ed permeate.
