1.2.1.4 Glass Funnels, 16306/16307

– Insert the base of the glass funnel
into the 17174 (17005) or
17175 (17006) silicone stopper
and insert the stopper into the
opening of the filtrate flask.
– Seat the white filter support into
the base.
– Remove a membrane filter from the
package, and centre it on the filter
support. For further instructions
on how to perform the analysis,
please refer to Section 3
"Microbiology" and/or Section 4
"Analyses."
– You can now easily and safely
connect the upper part and base
of the glass funnel using the
metal clamp.
2. Assembling the Entire
Vacuum Filtration System
The vacuum filtration systems from
Sartorius have been specially
matched to one another. Problems
may be encountered if components of
products from other manufacturers are
used with Sartorius systems.
Applications technology specialists
have performed numerous tests to
select ideal components and
materials which guarantee reliable
vacuum filtration.
The warranty will only be honoured
if the specified procedure ist followed
before, during and after vacuum
filtration.

– Use the vacuum hose to connect
the filtrate flask to the water trap or
barrier (Woulff's bottle or
Vacusart
). Before connecting the
®
vacuum hose, attach the safety
connector to the side outlet of the
5- and/or 2-liter filtrate flask (does
not apply to the 16606 1-liter
filtrate flask).
– Connect the vacuum pump to the
water trap. When installing the
®
Vacusart
, make sure that the
filtrate overflow spills back into the
suction flask and is not left to stand.
– Place the stopper onto the filtrate
flask, and connect the system
components as shown in the
drawings and descriptions under
1.1 "Multiple-Branch Systems"
and/or 1.2 "Individual Filter
Holder Systems."

3. Microbiological Tests

Described below is the
methodology to determine colony
counts with reusable systems
made of stainless steel, glass or
polycarbonate. Detailed description
of microbiological test methods used
for ready-to-connect units, such as
BioSart 100 Monitors or
250 Funnels, can found in the
Instructions for use accompanying the
corresponding product.
3.1 All system components coming
into contact with the sample and filter
membrane must be sterile. Sterility
can be ensured in different ways
depending on the materials utilised.
a) Autoclaving (for stainless steel,
glass and polycarbonate):
Assemble the unit so that the filter
membrane is clamped securely
between the base and the funnel.
The filter should not be clamped
too tightly; otherwise steam
will not penetrate the clamped
region of the membrane.
Adjust the three-way valve to the
12 o'clock position (does not
apply to individual glass filter
holders). It is not imperative to
sterilize the 16842 and/or
16843 stainless steel manifolds
when the adapted filter holders
(BioSart 100 or 250, stainless
steel, glass, polycarbonate) are
changed under sterile conditions.
Cover the inlets and outlets with
aluminium foil, and autoclave
according to the specifications of
the system components. After
sterilization, assemble the filtration
funnel, ascertaining all connections
are tight, and attach the funnel to
other system components.
Adjust the three-way valve to the
9 o'clock position.
b) Flaming (only for stainless steel):
Flaming prevents long waiting
periods (autoclave cycles) between
filtration runs and is described in
detail below.
3.2 Clean the laboratory bench, and
disinfect with alcohol or a commercial
disinfectant.
