KNAUER OQ documents are especially useful when performing
routine checks of KNAUER instruments. OQ documents are available
from your local dealer or via download from www.Knauer.net.An
automated OQ-check is available with KNAUER ChromGate
(some versions starting from V3.1 on).
The deuterium lamp used with the Smartline PDA Detector 2800 has an
extended lifetime to ensure long-lasting functionality and reliable
measurements with low noise and baseline drift as well as high
sensitivity. The actual condition of the lamp depends on different factors,
such as the number of lamp starts, the average burning time and the
user's noise and sensitivity requirements.
To check the functionality of the lamp, flush the flow cell with pure
methanol and determine the integration time in the chromatography
software (range: 200-400 nm with D
On delivery of the detector, the integration time should be 80 ms or less.
We recommend that the integration time be checked at regular intervals
under the conditions described, especially in cases where higher noise
levels or decreased sensitivity are observed when working with the
Smartline PDA Detector 2800. If these observations coincide with an
integration time of approx. 160 or higher, a new deuterium lamp should
be installed (see 'Changing the Lamp' on page 28).
To perform the wavelength accuracy test the KNAUER Smartline PDA
Detector 2800 needs to be equipped with an optionally available holmium
oxide filter test cell.
Holmium oxide glass has an absorption spectrum with characteristic
absorption maxima. By comparing the positions of the detected maxima
with the values in the enclosed holmium filter certificate the detector's
wavelength accuracy can be determined.
For checking wavelength accuracy of the PDA Detector 2800 according
to SOP 8, an absorption spectrum is recorded with the holmium oxide
filter moved into the optical path.

Wavelength Accuracy Check

1. Install the holmium filter cell as measuring cell.
2. Set the following parameters in the software:

3. Record an AutoZero spectrum. In order to do this the holmium
filter must be withdrawn from the holmium filter cell.
4. Push the holmium filter back into the cell and record the
absorption spectrum.
