Short Manual peqSTAR thermal cyclers
FlexGradient Technology™:
The temperature of each of the eight rows of the block is individually controlled! For the first time you can choose
either the perfectly linear temperature gradient – ideally suited for PCR optimizations, or independent lane control
– ideally suited for the use of different primer pairs in one PCR run.
First, the temperature range is defined by the buttons
'Center' and 'Gradient ±'. The standard gradient is
always realized from the top to the button over the 8
rows of the block. Moreover with the peqSTAR 96X you
have the opportunity to set a 16-gradient. In this case
different temperatures are realized in the left and right
part of the thermoblock.
As in this case you will also have a gradient in the
lanes from left to right, the temperatures displayed in
the software are only realized in the rows 3 and 10.
By selecting one of the temperature buttons
different options will be available: When the checkbox
was deactivated before, the single
temperatures of each row can be adjusted within a
special range using the button 'Adjust Temperature'.
Moreover the selected temperature can be used as new
center value or the gradient step can be changed in a
temperature step using the selected temperature.
Program options:
When setting up a new program, when saving or during programming (via the Info button) the program options
can be set. The following options are available for the user.

If a power failure had happened while a program was running the thermal cycler restarts the program at the point it was
stopped as soon as the power returns.
If the option 'Power Fail Denaturation' is activated the thermal block will heat to the stated temperature for the set time before
continuing with the program.

Using this function a global ramp rate in °C/s can be programmed. This global ramp rate will be used in all temperature and
gradient steps of the PCR program, but could be changed in a single temperature or gradient step manually by entering the
desired ramp value at the step.

Using this option you can decide whether in a gradient step the plateaus (Plateaus synchronized) or the Ramps (Ramps
synchronized) should be synchronized for all rows.

Via tube control you have the possibility to adjust temperature regulation for a temperature step according to sample volume. If
necessary, the temperature regulation will perform short term deliberate overshoots and undershoots depending on the entered
sample volume during PCR run.
Especially if combined with the emulation mode this function can be helpful if the emulated cycler used the tube control function
before, too.
